Изучить мембранный потенциал митохондрий, показатели ферментативной и неферментативной антиоксидантных систем, показатель повреждения мтДНК в митохондриях клеток колоректального рака (КРР) в зависимости от его расположения, наличия метастазов в лимфатических узлах и пола больных.</p></sec><sec><title>Пациенты и методы</title><p>Пациенты и методы. В исследование включены 132 пациента (52 женщины и 80 мужчин), у которых диагностирован рак толстой кишки T2-3N0-1M0. Митохондрии из клеток тканей кишки и опухоли выделяли методом дифференциального центрифугирования и определяли концентрацию супероксиддисмутазы (СОД-2), 8-оксо-7,8-дигидрогуанин (8OHDG), витаминов А и Е, глутатионпероксидазы (ГПО-1), мембранный потенциал (Δψm). Для статистической обработки результатов была использована программа Statistica 10.0.</p></sec><sec><title>Результаты</title><p>Результаты. В митохондриях опухоли прямой кишки с метастазами в лимфатических узлах у пациентов обоего пола увеличивалось содержание 8ОHDG, снижался Δψm относительно показателя в митохондриях опухоли больных без метастазов: у мужчин в 2,2 раза и 1,5 раза (р &lt; 0,05), у женщин в 1,3 раза (р &lt; 0,05) и 2,0 раза. Левая половина ободочной кишки: в митохондриях опухоли у мужчин с метастазами уровень 8ОHDG выше в 2,4 раза; Δψm, витамины А и Е снижены в 1,7 (р &lt; 0,05), 2,4 и 3,1 раза соответственно. В митохондриях опухоли женщин с метастазами уровень витаминов А и Е снижен в 1,7 (р &lt; 0,05) и 4,3 раза. Правая половина ободочной кишки: в митохондриях опухоли мужчин и женщин с метастазами в лимфатических узлах относительно показателей в митохондриях опухоли без метастазов были выше витамин А в 1,7 (р &lt; 0,05) и 2,4 раза и витамин Е в 4,2 и 3,9 раза. Уровень СОД-2 был ниже в 2,2 раза только в митохондриях опухоли с метастазами у мужчин.</p></sec><sec><title>Заключение</title><p>Заключение. Митохондриальный окислительный стресс представляет собой важное системное явление при прогрессирующем КРР, и его проявления зависят от пола и локализации.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Purpose of the study</title><p>Purpose of the study. To study the mitochondrial membrane potential, indicators of the enzymatic and non-enzymatic antioxidant systems, and the mtDNA damage index in the mitochondria of colorectal cancer (CRC) cells depending on its location, the presence of metastases in the lymph nodes, and the patient sex.</p></sec><sec><title>Patients and methods</title><p>Patients and methods. The study included results obtained from 132 patients (52 women and 80 men) with T2-3N0M0 colon cancer. Mitochondria from intestinal and tumor tissue cells were isolated using differential centrifugation. The concentration of SOD-2, 8OHdG, vitamins A and E, GPO-1, and the membrane potential (ΔψM) were determined in mitochondria. Statistical analysis was performed using the program Statistica 10.0.</p></sec><sec><title>Results</title><p>Results. In rectal tumor mitochondria with lymph node metastases, both men and women demonstrated increased 8-OHdG levels and decreased ΔψM compared with patients without lymph node metastases: in men, by 2.2-fold and 1.5-fold (p &lt; 0.05), and in women, by 1.3-fold (p &lt; 0.05) and 2.0-fold, respectively. In the left colon, tumor mitochondria in men with metastases showed a 2.4-fold increase in 8-OHdG, while ΔψM and vitamins A and E levels were reduced by 1.7-fold (p &lt; 0.05), 2.4-fold, and 3.1-fold, respectively. In women, lymph node metastases were associated with decreased levels of vitamins A and E by 1.7-fold (p &lt; 0.05) and 4.3-fold. In the right colon, tumor mitochondria in patients with metastases showed higher levels of vitamins A and E compared with those without metastases: vitamin A increased by 1.7-fold (p &lt; 0.05) in men and 2.4-fold in women, and vitamin E by 4.2-fold and 3.9-fold, respectively. SOD-2 levels were 2.2-fold lower only in tumor mitochondria with metastases in men.</p></sec><sec><title>Conclusion</title><p>Conclusion.
