А.Ф. Цыба были синтезированы водорастворимые диалкиламиноалкиловые эфиры индол‑3‑карбоновой кислоты с высокой иммуномодулирующей, противовирусной и противоопухолевой активностью. С целью усиления противоопухолевого действия в структуру соединений добавлены бром- или бис-(2‑хлорэтил)амино-заместители, придающие молекулам дополнительные цитотоксические алкилирующие свойства. Нами успешно синтезированы соединения – T1089 и T1167.</p><sec><title>Цель исследования</title><p>Цель исследования. Изучить противоопухолевые и антиметастатические свойства бром-, бис-(2‑хлорэтил)амино-замещенных производных N-метильных этиловых эфиров индол‑3‑карбоновой кислоты (T1089 и T1167).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Исследование проводили на 65 мышах с подкожно трансплантированной саркомой S‑37 (9 × 105 клеток/мышь). Соединения вводили внутрибрюшинно: Т1089–40 мг/кг, Т1167 в дозе 30 мг/кг. Противоопухолевый эффект оценивали по динамике роста первичных узлов, морфометрическим и гистологическим показателям.</p></sec><sec><title>Результаты</title><p>Результаты. Исследуемые соединения вызывали торможение роста первичного узла саркомы S‑37 (ТРО Т1089– до 22,4 %, ТРО Т1167 – до 30,5 %), повышали содержание дистрофических и некротических изменений (Т1089 – на 36 %, Т1167 – на 55 %) в опухоли, снижали митотический индекс в 2 раза, проявляли антиметастатическую активность, уменьшая метастатические области в паховых (10‑е сутки: Т1089 – на 76 %, Т1167 – на 89 %) и подвздошных лимфоузлах (10‑е сутки: Т1089–28,7 %, Т1167 – на 60,8 %; 16‑е сутки: Т1089 – на 53,3 %, Т1167–45,6 %.</p></sec><sec><title>Заключение</title><p>Заключение. Показана противоопухолевая и антиметастатическая активность соединений Т1089 и Т1167 на модели саркомы S‑37. Полученные данные свидетельствуют о перспективности соединений, сочетающих алкилирующую цитотоксичность с присущим данному классу иммуномодулирующим потенциалом. Дальнейшие исследования предполагают доклиническую разработку соединений, включающую оценку безопасности, расширение спектра изучаемых опухолевых моделей и детальное изучение механизмов действия.</p></sec></abstract><trans-abstract xml:lang="en"><p>The Laboratory of Radiation Pharmacology has previously developed water-­soluble dialkylaminoalkyl esters of indole‑3‑carboxylic acid, which demonstrated high immunomodulatory, antiviral, and antitumor activity. Based on these established properties, we aimed to enhance their antitumor effect by incorporating cytotoxic alkylating bromo- or bis(2‑chloroethyl)amino substituents into their molecular structure. Compounds T1167 and T1089 were successfully synthesized.</p><sec><title>Purpose of the study</title><p>Purpose of the study. To investigate the antitumor and antimetastatic properties of bromo-, bis(2‑chloroethyl)amino-substituted derivatives of N-methyl ethyl esters of indole‑3‑carboxylic acid (T1089 and T1167).</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The study was performed on 65 mice with subcutaneously transplanted lymphogenously metastasizing sarcoma S‑37 (9 × 105 cells/mouse). Compounds were administered intraperitoneally: T1089 at 40 mg/kg, T1167 at 30 mg/kg. Antitumor efficacy was assessed by primary tumor growth dynamics, morphometric and histological parameters.</p></sec><sec><title>Results</title><p>Results. The studied compounds inhibited primary tumor growth of sarcoma S‑37 (TGI for T1089–22,4 %, for T1167–30,5 %), increased dystrophic and necrotic changes in tumors (T1089 – by 36 %, T1167 – by 55 %), reduced mitotic index by 2‑fold, and exhibited antimetastatic activity, decreasing metastatic areas in inguinal lymph nodes (day 10: T1089 – by 76 %, T1167 – by 89 %) and iliac lymph nodes (day 10: T1089 – by 28,7 %, T1167 – by 60,8 %; day 16: T1089 – by 53,3 %, T1167 – by 45,6 %).</p></sec><sec><title>Conclusion</title><p>Conclusion. Antitumor and antimetastatic activity of compounds T1089 and T1167 was demonstrated in the sarcoma S‑37 model. The obtained data indicate the promise of compounds combining alkylating cytotoxicity with the immunomodulatory potential inherent to this class.
